Definition
Amlodipine is the besylate salt of amlodipine, a drug belonging to a class of drugs known as dihydropyridine calcium channel blockers. Approved by the FDA in 1987, it has antihypertensive and antianginal effects. Dihydropyridine calcium channel blockers selectively act on myocardial and peripheral blood vessels, resulting in dilation of arteries.
Therefore, the drug is used for the treatment of hypertension and different types of angina.

6.Mechanism of action of Amlodipine Besylate
The influx of calcium ions is needed for the contractility of cardiac muscle cells and vascular smooth muscle cells. The besylate salt of amlodipine is a dihydropyridine calcium antagonist. Therefore it inhibits the influx of calcium ions into these cells by blocking the calcium channel. The effect is greater on vascular smooth muscle cells than cardiac muscle cells, and consequently, peripheral vascular resistance and blood pressure are decreased.

Phtaloyl amlodipine is the starting material used by the manufacturer to prepare amlodipine and its salts. It is processed with aqueous solutions of primary amines in water-immiscible solvents such as methylene chloride, ethylene dichloride, chloroform ethyl acetate, and toluene. This is known as the deprotection reaction. Afterward, it is treated with benzene sulfonic acid, and the final product is obtained.

Treat high blood pressure
By selectively blocking the calcium ion influx across cell membranes, it dilates the main coronary and systemic arteries. It leads to a reduction in peripheral vascular resistance, which in turn decreases blood pressure.
Chest pain
The drug is used to treat angina, which is chest pain. It dilates peripheral arterioles, reducing total peripheral resistance (afterload) against which the heart works. This leads to a decrease in the myocardial oxygen demand and alleviates exertional angina.
The arteriolar dilation also increases oxygen delivery to the heart in patients experiencing a coronary artery spasm, making the drug valuable in the case of Prinzmetal's or variant angina.
9.Amlodipine Besylate dosage
The oral antihypertensive dose for adults is 5 mg once daily, with the maximum being 10 mg daily. For older patients, it is started at 2.5 mg once daily.
With regards to angina and coronary artery disease, the dose is 5-10 mg once daily.

The side effects include swelling in the legs and ankles, stomachache and nausea, dizziness, flushing, arrhythmias, palpitations, muscle rigidity, and tremors. However, these are usually mild in nature.
